Abstract
Tissue culture technique could produce plenty and high quality seedlings. One obstacle in tissue culture of banana cv tanduk is non syncronized shoots. Thus this study aimed to obtain optimal medium for in vitro shoot multiplication and elongation. The research was conducted in Tissue Culture Laboratory in Citayam Experimental Station, Indonesian Center for Agricultural Biotechnology and Genetic Resources Research and Development from October 2020 to January 2021. In vitro shoots after 12 times of subculture were used as the explants. There were six treatments: HM1 (MS + BA 5 μM + IAA 0 μM), HM2 (MS + BA 10 μM + IAA 0 μM), HM3 (MS + BA 5 μM + IAA 1 μM), HM4 (MS + BA 10 μM + IAA 1 μM), HM5 (MS + BA 5 μM + IAA 4 μM), HM6 (MS + BA 10 μM + IAA 4 μM) and hormon-free MS as the control. Each treatment was replicated 7 times. The survival rate, number of shoots nodule, number of shoots, number of leaves, number of roots, length of roots, and length of shoots were observed at 4 and 8 weeks after culture. The result showed that the best culture medium for shoot multiplication was HM4 and the best treatment for shoot elongation was hormon-free MS medium.
