Abstract
The development of drought-tolerant alfalfa will support the livestock’s productivity and health, soil and environmental health and conservation. One of the main problems in increasing livestock productivity is the provision of sustainable feeds, especially in dry areas. Alfalfa (Medicago sativa) is one of nutritious forage. In addition, it is also widely used in the pharmaceutical, food supplements, and soil fertilizer. In order to develop drought-tolerant alfalfa, induced mutation program was carried out. Induced mutation was targeted to embryogenic callus of 3 Alfalfa varieties using UV-C irradiation at 254 nm, intensity 29.13 mol photon/m2/s for 30 and 60 minutes irradiation. In vitro selection of irradiated alfalfa callus was done using MS media (Murashige & Skoog) containing 10%, 20% and 30% of 6000 MW polyethylene glycol (PEG). The survival rate of somatic embryos in 30% PEG media for alfalfa V1, with UV-C irradiation for 30 and 60 minutes were 55% and 67% respectively, for V2 were 47% and 46%, and for V3 were 42% and 40%. In order to obtain the best promising clones of drought-tolerant Alfalfa, further field testing is necessary for these in vitro selected clones.
