Abstract
Salinity is a major obstacle in maintaining the growth and development of rice plants grown in the coast areas. Superior varieties with saline tolerance are needed to overcome this problem. The aim of this research is to obtain LD50 of Inpari 42 Agritan GSR lowland rice seeds, induce mutant callus from irradiated seeds, and obtain LC50 NaCl value in salinity tolerant mutant callus. Determining LD50 of rice seeds was carried out using a Complete Randomized Design (CRD) 1 factor, consisted of 13 irradiation doses: 0, 100, 200, 300, 400, 500, 600, 700, 700, 800, 900, 1000, 1100, and 1200 Gy. The results showed that the LD50 value of Inpari 42 Agritan GSR rice seeds is 670.65 Gy. Induction of Inpari 42 Agritan GSR rice mutant callus was done using a completely randomized design (CRD) 1 factor, consisted of 4 levels consentration of 2,4-D: 0, 1, 3, and 5 mg L-1. The results of the experiment showed that the addition of 2,4-D has a very significant effect on the percentage and diameter of callus. On MS media without agreeing 2,4-D callus was not formed (0%) until week 14. While in different 2,4-D agreement (1, 3, and 5 mg L-1) mutant callus formed with 58, 61, and 50%. Selection of mutant callus to obtain LC50 values was conducted using a completely randomized design (CRD) 1 factor, consisted of 3 concentration level of NaCl; 100, 150, and 200 mM. LC50 value of NaCl was obtained at 88.61 mM.
