Abstract
Medium is one of the important factors in determining the growth quality of shallot explants in vitro. This research aims to determine the best media formulation for shoot multiplication and micro bulb formation in shallots. The results of the shoot multiplication experiment showed that giving Plant Growth Regulator (PGR) had a very significant and significant effect on the number of shoots, number of leaves, number of roots, and root length and had no significant effect on leaf length for 6 weeks after culture in vitro. The difference in the concentration of paclobutrazol and sugar did not significantly affect the number of tubers formed and tuber weight for 5 weeks after culture in vitro in the micro bulb formation research from this research, it was found that the best media for shoot multiplication was media treatment ((Murashige and Skoog (MS) + kinetin 2 mg/l + Naphthalene Acetic Acid (NAA) 2 mg/l + casein hydrolyzate 500 mg/l) which produced the highest number of shoots, namely 11.3 and the best medium for micro bulb formation is the media treatment (MS + paclobutrazol 0.4 mg/l + sugar 90 g/l) with the highest number of micro bulb formation 1.43 bulbs.
