- ItemAnalisis Keragaman Genetik Aksesi Ubi Jalar Lokal Menggunakan Marka Simple Sequence Repeat (SSR)(KOMISI NASIONAL SUMBER DAYA GENETIK, 2021-09-15) Kurniawan ...[at al], Hakim; Balai Besar Penelitian dan Pengembangan Bioteknologi dan Sumberdaya Genetik PertanianIndonesia is the second center of origin after South America for sweet potato (Ipomoea batatas (L.) Lamb.) genetic resources, with the highest diversity found in Papua. This commodity is widely cultivated and uses both for food and industrial raw materials in various regions of Indonesia. The objective of this research is to study genetic diversity of sweet potato accessions collected from various areas in Indonesia. A total of 90 accessions of local sweet potatoes collected from Java, Nusa Tenggara, Sulawesi and Papua were used in this study. DNA was extracted and isolated for further molecular analysis using 18 SSR markers. The results showed that of the 18 SSR markers used, 13 markers (72%) were highly informative (PIC>0.73). From the results of the analysis, the average value of the major allele frequency, the number of alleles, gene diversity, heterozygosity and polymorphic information content/PIC were 0.32, 13.7, 0.79, 0.38, and 0.77, respectively. The IBSSR19 marker is known to be the most productive and informative marker because it is able to display the highest number of alleles (27 alleles), the highest gene diversity (0.92) as well as the highest PIC value (0.91). The result of cluster analysis showed that 90 accessions of sweet potato were separated into three groups, each consisting of 32, 46 and 12 accessions. The second group is further separated into two sub-groups, each consists of 20 and 26 accessions. This grouping does not reflect with the provenance of the accessions.

H. Kurniawan and L. Gunarto. Microalgae mass cultivation process has become an interesting issue these days due to the beneficial values and industrial application of these microorganisms. Products of microalgae have been very useful to human needs in various forms, such as food stuffs (essential fatty acids, essential amino acids, vitamins, and food colors), enzymes (proteases, phosphogiyceric kinases, amino acid oxidases, superoxide dismutases, and restriction endonucieases) as well as pharmaceutical and therapeutically agents (antibiotics, anticancer, antiviral, toxins, inhibitory enzymes, and hypocholesterolemic effects). Currently, mass production of microalgae is being done through heterotrophic and photoheterotrophic cultures either outdoors on ponds or indoor using both enclosed bioreactors and photobioreactors. These techniques could be improved by the application of biotechnology, such as use of cell immobilization technique. This technique could improve productivity of the micoalgae mass production system.

- ItemPengembangan Sistem Pangkalan Data (Database) Plasma Nutfah Tanaman Pangan(BB Biogen, 2005) Kurniawan ...[at al], Hakim; Balai Besar Penelitian dan Pengembangan Bioteknologi dan Sumberdaya Genetik PertanianKegiatan pengelolaan plasma nutfah tanaman pangan melibatkan cukup banyak kegiatan mulai dari eksplorasi/introduksi, registrasi, konservasi, karakterisasi, evaluasi hingga pemanfaatan plasma nutfah yang bersangkutan bagi kegiatan penelitian pemuliaan. Pada setiap kegiatan tersebut dihasilkan banyak sekali data dan informasi penting yang harus didokumentasikan. Perkembangan peningkatan aktivitas bank gen dari waktu ke waktu menuntut tersedianya sistem basis data dan informasi yang dapat diakses secara mudah, cepat dan akurat. Sejak tahun 2000 telah dilakukan penyusunan sistem database plasma nutfah tanaman pangan yang dikelola menggunakan Microsoft Access. Sampai dengan Februari 2005, database plasma nutfah tanaman pangan telah menampung sebanyak 9.423 record dari 16 komoditas yang meliputi 3.653 aksesi padi (41 deskriptor), 734 aksesi jagung (29 deskriptor), 210 aksesi sorgum (22 deskriptor), 771 aksesi kedelai (28 deskriptor), 619 aksesi kacang tanah (19 deskriptor), 1.024 aksesi kacang hijau (20 deskriptor), 434 aksesi ubi kayu (23 deskriptor), 1.426 aksesi ubi jalar (35 deskriptor), 112 aksesi kacang tunggak (22 deskriptor), 34 aksesi ubi kelapa (5 deskriptor), 33 aksesi gembili (5 deskriptor), 48 aksesi ganyong (5 deskriptor), 115 aksesi talas (5 deskriptor), 58 aksesi belitung (5 deskriptor), 88 aksesi padi liar (5 deskriptor), dan 64 aksesi terigu (5 deskriptor). Informasi yang dihasilkan selanjutnya dihimpun dan dicetak dalam bentuk katalog plasma nutfah tanaman pangan yang senantiasa diperbarui setiap tahun. Diharapkan katalog plasma nutfah tanaman pangan yang dihasilkan akan dapat disebarluaskan ke berbagai kalangan pengguna. Saat ini juga telah dikembangkan penyusunan katalog plasma nutfah tanaman pangan dalam compact disc serta sedang dikembangkan sistem database berbasis web untuk memudahkan akses data secara langsung bagi para pengguna melalui jaringan internet.
